tubeless tires?
There are companies that make spoked wheels that are sealed so you can use a tubeless tire w/o an intertube. The wheel / spokes are all laced up and they apply a rubberized goo like a aircraft gas tank sealer over the tire-side of the nipples and then lock the wheel in a chamber that allows them to pull a vacuum on the spoke-side of the wheel. This pulls the rubberized sealer in around the nipples sealing the rim / spoke / spoke nipple interfaces.
Just think what a pain it'll be to do that and repair it later if leak develops.

You could try sealing the spoke holes, but then your tubeless tire would not have the safety benefit of a bead retention shoulder on the rim.
