If you are going to bother, why not fit two? Put the two rim locks almost opposite each other, 90deg off the valve stem. Putting one rim lock a spoke off of exactly opposite will offset the weight of the valve stem. I did this, and only needed a .5oz weight to balance the front wheel. And with twin rimlocks, you can ride a flat without the bead breaking loose.

Totally depends on your tire. GP110 has a pliable carcass compared to a D606, but with rimlocks you could go to 12 or 10psi and improve your soft-terrain traction.

I've cracked and dented a stock KLR front rim without getting a pinch flat. The heavy tubes are great.

I run the maximum pressure for the highway.

Mike, I have had my front GP110 mounted backwards for over 9000 miles with no ill handling effects or wear issues (thanks for the tip Jake). I think I have another 6 to 7000 miles left before it hits the wear marks. It handles ok off-road when aired down but watch yourself in sand or mud. Nice road manners with the cool Stuka JU-87 dive bomber howl everyone mentions when leaned over. I use 33 psi for the road and 18 to 20psi for off-road (no rim locks).
